Purity/Purification
Greater than 99.0% as determined by: (a) Analysis by Gel Filtration. (b) Analysis by SDS-PAGE.
Form/Format
The protein was lyophilized from a concentrated (1mg/ml) solution with 0.02-0.03% NaHCO3.
Sterile Filtered White lyophilized (freeze-dried) powder.
Sequence
The sequence of the first six N-terminal amino acids was determined and was found to be Ala-Val-Gln-Thr-Val-Pro
Solubility
It is recommended to reconstitute the lyophilized Placental Lactogen in sterile water or 0.4% NaHCO3 adjusted to pH 8-9, not less than 100 ug/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein.
Protein Content
UV spectroscopy at 280 nm using the absorbency value of 0.73 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
Biological Activity
Placental Lactogen Human is biologically active as evidenced by inducing proliferation of Nb2 cells.
Preparation and Storage
Lyophilized Placental Lactogen Human Recombinant although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution Placental Lactogen should be stored at 4 degree C between 2-7 days and for future use below -18 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.
Related Product Information for CSH1 active protein
Description: Placental Lactogen Human Recombinant, is a single polypeptide chain containing 199 amino acids and an additional Ala at the N-terminus having a molecular mass of approximately 22.4 kDa. Placental Lactogen Recombinant is purified by proprietary chromatographic techniques.

Introduction: Placental Lactogen is a polypeptide hormone that is produced by the Syncytiotrophoblasts of the Placenta, also known as chorionic somatomammotropin. It has both Growth Hormone and Prolactin activities on growth, lactation, and luteal steroid production. In women, placental lactogen secretion begins soon after implantation and increases to 1 g or more a day in late pregnancy. Placental lactogen is also an insulin antagonist.Placental Lactogen Bovine is also capable of activating human and other heterologous GH receptors but not ruminat GH receptors.

NCBI Description

The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones and plays an important role in growth control. The gene is located at the growth hormone locus on chromosome 17 along with four other related genes in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. Although the five genes share a remarkably high degree of sequence identity, they are expressed selectively in different tissues. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed mainly in the placenta and utilizes multiple transcription initiation sites. Expression of the identical mature proteins for chorionic somatomammotropin hormones 1 and 2 is upregulated during development, although the ratio of 1 to 2 increases by term. Mutations in this gene result in placental lactogen deficiency and Silver-Russell syndrome. [provided by RefSeq, Jul 2008]

Uniprot Description

CSH1: Produced only during pregnancy and is involved in stimulating lactation, fetal growth and metabolism. Does not interact with GHR but only activates PRLR through zinc-induced dimerization. Belongs to the somatotropin/prolactin family.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 17q24.2

Cellular Component: extracellular region

Molecular Function: metal ion binding; hormone activity

Product Notes

The CSH1 (Catalog #AAA143698) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first six N-terminal amino acids was determined and was found to be Ala-Val-Gl n-Thr-Val- Pro. It is sometimes possible for the material contained within the vial of "Placental Lactogen, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
