For Research Use Only!
Host
E Coli
Purity/Purification
Highly Purified
98% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Form/Format
Supplied as a lyophilized powder. No additives.
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Ser-Val-Val-Thr.
Biological Activity
GRO-gamma is fully biologically active when compared to standard. The Biological activity is calculated by its ability to chemoattract CXCR2 transfected 293 cells using 10.0-100.0 ng/ml.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of Human GRO-gamma.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm.
2. Analysis by RP-HPLC, using a calibrated solution of Human GRO-gamma as a Reference Standard.
Reconstitution
Reconstitute the lyophilized Human GRO-gamma in sterile 18MO-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for GRO gamma active protein
CXCL3/GROg is a small cytokine belonging to the CXC chemokine family. CXCL3 controls migration and adhesion of monocytes and mediates it effects on its target cell by interacting with a cell surface chemokine receptor called CXCR2. The three GRO cDNAs encode 107 amino acid precursor proteins from which the N-terminal 34 amino acid residues are cleaved to generate the mature GROs. There are no potential N-linked glycosylation sites in the amino acid sequences. GRO expression is inducible by serum or PDGF and/or by a variety of inflammatory mediators, such as IL-1 and TNF, in monocytes, fibroblasts, melanocytes and epithelial cells. In certain tumor cell lines, GRO is expressed constitutively.Similar to other alpha chemokines, the three GRO proteins are potent neutrophil attractants and activators. In addition, these chemokines are also active toward basophils. All three GROs can bind with high affinity to the IL-8 receptor type B.

GRO gamma Recombinant, Human is a 7.9kD protein containing 73 amino acid residues.

NCBI and Uniprot Product Information

NCBI GI #
NCBI Official Full Name
GRO-gamma

NCBI Description

This antimicrobial gene encodes a member of the CXC subfamily of chemokines. The encoded protein is a secreted growth factor that signals through the G-protein coupled receptor, CXC receptor 2. This protein plays a role in inflammation and as a chemoattractant for neutrophils. [provided by RefSeq, Sep 2014]

Uniprot Description

CXCL3: Ligand for CXCR2. Has chemotactic activity for neutrophils. May play a role in inflammation and exert its effects on endothelial cells in an autocrine fashion. In vitro, the processed form GRO-gamma(5-73) shows a fivefold higher chemotactic activity for neutrophilic granulocytes. Belongs to the intercrine alpha (chemokine CxC) family.

Protein type: Secreted, signal peptide; Secreted; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 4q21

Cellular Component: extracellular space; extracellular region

Molecular Function: chemokine activity; CXCR chemokine receptor binding

Biological Process: neutrophil chemotaxis; G-protein coupled receptor protein signaling pathway; response to lipopolysaccharide; immune response; positive regulation of leukocyte chemotaxis; inflammatory response; regulation of cell proliferation

Product Notes

The GRO gamma (Catalog #AAA650156) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Ser-Va l-Val-Thr. It is sometimes possible for the material contained within the vial of "GRO gamma, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
