Intended Uses: The TX test kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring TX in chicken biological samples. If the ELISA is to be used outside the intended use, the user may need to optimize for said use.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the ovotransferrin (TX) present in samples reacts with the anti-TX antibodies, which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-TX antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound TX. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of TX in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of TX in the test sample. The quantity of TX in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Uniprot Description

Function: Transferrins are iron binding transport proteins which can bind two Fe3+ ions in association with the binding of an anion, usually bicarbonate. It is responsible for the transport of iron from sites of absorption and heme degradation to those of storage and utilization. Serum transferrin may also have a further role in stimulating cell proliferation.Transferrin has a bacteriostatic function. Its concentration in avian egg is the highest concentration of any transferrin in vivo. Transferrin in liver is regulated by the iron levels but not in the oviduct. These two forms of transferrin (liver and oviduct) differ only by their carbohydrate composition.
