Application Notes
ELISA, Ouchterlony, Immunoblotting. Used in vitro differentiation of mouse ES cells into neural cells. Suitable for use in mouse embryonic stem cell research. Use in Basal Medium A and as a coating agent for coverslips. This component is recommended for growth and survival of neural cell types during in vitro differentiation of mouse ES cells.

Measured by its ability to support cell attachment and spreading when used as a substratum for cell culture.
Endotoxin Level
1.0 EU per 1 ug of fibronectin as determined by the LAL method.
Recommended Dilution
To coat coverslips, prepare a 1mg/ml solution. For use as a substratum for cell attachment and spreading, use at 1-5ug/cm2. To support cell spreading, add 0.5-50ug/ml in media.
Preparation and Storage
Samples are stable for up to six months at 2-8 degree C. Product is not stable at room temperature. Avoid vortexing and excessive agitation. Aliquot into 50-100ul volumes and store at -20 degree C for up to six months in a manual defrost
freezer. Avoid repeated freeze-thaw cycles.

Fibronectin is an extracellular matrix (ECM) component and is one of the primary cell adhesion molecules.1 It is composed of multiple homologous repeats and contains many functional domains. The occurrence of different isoforms is due to alternative mRNA splicing of the ED-A, ED-B and III-CS regions and subsequent post-translational modification. Uniprot Description

Function: Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape. May be involved in osteoblast compaction

Tissue specificity: In early Xenopus embryo, cellular forms of fibronectin predominate which include both extra domains. In fibronectin of embryonic and adult liver the connecting strand 3 can be either completely excluded or included.

Post-translational modification: Forms covalent cross-links mediated by a transglutaminase, such as F13A or TGM2, between a glutamine and the epsilon-amino group of a lysine residue, forming homopolymers and heteropolymers (e.g. fibrinogen-fibronectin, collagen-fibronectin heteropolymers).
