Principle of the Assay: The Mouse CD166 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD166 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse CD166 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD166 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD166 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD166 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: CD166 antigen is a 100-105 kD typeI transmembrane glycoprotein that is a member of the immunoglobulin superfamily of proteins. In humans it is encoded by the ALCAM gene. The gene was mapped to human chromosome 3q13.1-q13.2 by fluorescence in situ hybridization of cDNA probes to metaphase chromosomes. It is also called CD166 (cluster of differentiation 166), MEMD, SC-1/DM-GRASP/BEN in the chicken, and KG-CAM in the rat. It is expressed on activated T cells, activated monocytes, epithelial cells, fibroblasts, neurons, melanoma cells, and also in sweat and sebaceous glands. CD166 protein expression is reported to be upregulated in a cell line deriving from a metastasizing melanoma. CD166 plays an important role in mediating adhesion interactions between thymic epithelial cells and CD6+ cells during intrathymic T cell development. Recently, CD166 has also been used as a potential cancer stem cell marker.

Uniprot Description

ALCAM: Cell adhesion molecule that binds to CD6. Involved in neurite extension by neurons via heterophilic and homophilic interactions. May play a role in the binding of T- and B-cells to activated leukocytes, as well as in interactions between cells of the nervous system. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell adhesion; Membrane protein, integral

Cellular Component: axon; cell soma; external side of plasma membrane; focal adhesion; immunological synapse; intrinsic to plasma membrane; T cell receptor complex

Molecular Function: protein binding

Biological Process: axon guidance; heterophilic cell adhesion; motor axon guidance

Product Notes

The Mouse ALCAM alcam (Catalog #AAA8248418) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8248418 ELISA Kit recognizes Mouse ALCAM. It is sometimes possible for the material contained within the vial of "CD166, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
