Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PRKAR2B elisa kit
Description: This Mouse CAMP-dependent protein kinase type II-beta regulatory subunit (PRKAR2B) ELISA Kit employs a two-site sandwich ELISA to quantitate PRKAR2B in samples. An antibody specific for PRKAR2B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PRKAR2B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRKAR2B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRKAR2B bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The inactive holoenzyme of AMPK is a tetramer composed of two regulatory and two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of regulatory subunits bound to four cAMP and two free monomeric catalytic subunits. Four different regulatory subunits and three catalytic subunits of AMPK have been identified in humans. PRKAR2b is one of the regulatory subunits. This subunit can be phosphorylated by the activated catalytic subunit. This subunit has been shown to interact with and suppress the transcriptional activity of the cAMP responsive element binding protein 1 (CREB1) in activated T cells. Knockout studies in mice suggest that this subunit may play an important role in regulating energy balance and adiposity.

Uniprot Description

PKAR2B: a regulatory subunit of cAMP-regulated protein kinase. The inactive form of the enzyme is composed of two regulatory chains and two catalytic chains. Activation by cAMP produces two active catalytic monomers and a regulatory dimer that binds four cAMP molecules. Four types of regulatory chains are found: I-alpha, I-beta, II-alpha, and II-beta. Their expression varies among tissues and is in some cases constitutive and in others inducible. Type II regulatory chains mediate membrane association by binding to anchoring proteins, including the MAP2 kinase.

Product Notes

The Mouse PRKAR2B prkar2b (Catalog #AAA9717277) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9717277 ELISA Kit recognizes Mouse PRKAR2B. It is sometimes possible for the material contained within the vial of "CAMP-Dependent Protein Kinase Type II-beta Regulatory Subunit (PRKAR2B), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
