* Allow all reagents to warm to room temperature for at least 30 minutes before opening.

Background: TRPM4 has 1,040 amino acids and contains 6 putative transmembrane domains. Northern blot analysis detected TRPM4 expression in most adult tissues tested, with highest levels in heart, prostate, and colon. In fetus, TRPM4 expression was most abundant in kidney. There were at least 2 distinct bands detected (6.2 and 4.2 kb), indicating alternative splicing of TRPM4. In addition, a smaller but relatively abundant 2.4-kb transcript was detected in testis.The shorter protein results from alternative splicing, with the third and fourth exons being spliced out and the beginning of the first exon being truncated. As a consequence, the first in-frame methionine lies in the fifth exon, resulting in the deletion of the first 174 amino acids.

Mediates transport of monovalent cations (Na+ > K+ > Cs+ > Li+), leading to depolarize the membrane. It thereby plays a central role in cadiomyocytes, neurons from entorhinal cortex, dorsal root and vomeronasal neurons, endocrine pancreas cells, kidney epithelial cells, cochlea hair cells etc. Participates in T-cell activation by modulating Ca2+ oscillations after T lymphocyte activation, which is required for NFAT-dependent IL2 production. Involved in myogenic constriction of cerebral arteries. Controls insulin secretion in pancreatic beta-cells. May also be involved in pacemaking or could cause irregular electrical activity under conditions of Ca2+ overload. Affects T-helper 1 (Th1) and T-helper 2 (Th2) cell motility and cytokine production through differential regulation of calcium signaling and NFATC1 localization. Enhances cell proliferation through up-regulation of the beta-catenin signaling pathway (). Essential for the migration but not the maturation of dendritic cells.
