Principle of the Assay||The Mouse ADAM12 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse ADAM12 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse ADAM12 coated on a 96-well plate. Standards and samples are pipetted into the wells and ADAM12 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse ADAM12 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ADAM12 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: ADAM12 (Disintegrin and metalloproteinase domain-containing protein 12), also known as MLTN, is an enzyme that in humans is encoded by the ADAM12 gene. This gene encodes a member of the ADAM (a disintegrin and metalloprotease) protein family. Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. This gene has two alternatively spliced transcripts: a shorter secreted form and a longer membrane-bound form. The shorter form is found to stimulate myogenesis. By RT-PCR and immunoblot analyses that expression of mouse Adam12 increases during muscle regeneration, while the levels of other ADAMs remain constant. Immunofluorescence analysis revealed staining of small, newly formed muscle fibers in regenerating but not normal adult muscle cells. By using of fluorescence in situ hybridization, the ADAM12 gene is mapped to human chromosome 10q26.3.

Uniprot Description

ADAM12: contains a disintegrin and metalloprotease (ADAM) domain 12, which is a member of the ADAM protein family. Involved in skeletal muscle regeneration, specifically at the onset of cell fusion, and the differentiation of macrophage-derived giant cells (MGC) and osteoclasts. 2 alternative splicing isoforms have been described: a shorter secreted form and a longer membrane-bound form. The shorter form stimulates myogenesis.

Product Notes

The Mouse ADAM12 adam12 (Catalog #AAA8244723) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244723 ELISA Kit recognizes Mouse ADAM12. AAA Biotech's ADAM12 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the ADAM12 adam12 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ADAM12, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
