Principle of the Assay||The Mouse CD144 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD144 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD144 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD144 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD144 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD144 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Cadherin 5, type 2 or VE-cadherin also known as CD144, is encoded by the gene CDH5. It is a classical cadherin from the cadherin superfamily. This gene is mapped to mouse chromosome 8. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of theorganization of the intercellular junctions. It has found that PECAM1, VE-cadherin and VEGFR2 together are sufficient to confer responsiveness to flow in heterologous cells.

NCBI Description

This gene encodes a member of the cadherin family of calcium-dependent glycoproteins that mediate cell adhesion and regulate many morphogenetic events during development. The encoded preproprotein is further processed to generate a mature protein. Mice lacking the encoded protein die in utero due to vascular insufficiency, caused by increased endothelial apoptosis. Multiple distinct genes of the cadherin family, including this gene, are found on chromosome 8. [provided by RefSeq, Oct 2015]

CDH5: Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. This cadherin may play a important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. It associates with alpha-catenin forming a link to the cytoskeleton. Acts in concert with KRIT1 to establish and maintain correct endothelial cell polarity and vascular lumen. These effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. Required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. Interacts via cadherin 5 domain with PTPRB. Interacts with TRPC4. Interacts with KRIT1. Endothelial tissues and brain.
