Acyl-CoA synthetase long-chain family member 4 (ACSL4), a member of the ACSL family, is a unique isozyme that preferentially catalyzes several polyunsaturated fatty acids (PUFAs) such as arachidonic acid (AA). ACSL4 is thought to be an important isozyme for PUFA metabolism. ACSL4 is involved in biological responses including inflammation, steroidogenesis, cell death, female fertility, and cancer. ACSL4 is an essential component for ferroptosis execution. ACSL4 inhibition is a viable therapeutic approach to preventing ferroptosis-related diseases. ACSL4 is not only a sensitive monitor of ferroptosis, but also an important contributor of ferroptosis.
