cGMP-dependent protein kinase II (cGKII; encoded by PRKG2) is a serine/threonine kinase that is critical for skeletal growth in mammals and plays multiple roles in cellular signaling pathways to control proliferation and differentiation; in mice, cGKII deficiency results in dwarfism. Using radiographic analysis, scientists determined this growth defect as a consequence of an elongated growth plate and impaired chondrocyte hypertrophy. The kinase activity of cGMP-dependent, type II, protein kinase (PRKG2) is required for the proliferative to the hypertrophic transition of growth plate chondrocytes during endochondral ossification. Loss of PRKG2 function in rodent and bovine models results in dwarfism. Cyclic GMP-dependent protein kinase II (cGKII; PRKG2) phosphorylates a variety of biological targets and has been identified as a gout-susceptible gene.
