Mouse Serine/threonine-protein kinase 4 (STK4) ELISA Kit
Principle of the Assay: STK4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-STK4 antibody and an STK4-HRP conjugate. The assay sample and buffer are incubated together with STK4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the STK4 concentration since STK4 from samples and STK4-HRP conjugate compete for the anti-STK4 antibody binding site. Since the number of sites is limited, as more sites are occupied by STK4 from the sample, fewer sites are left to bind STK4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The STK4 concentration in each sample is interpolated from this standard curve.

Key component of the Hippo signaling pathway which plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein STK3/MST2 and STK4/MST1, in complex with its regulatory protein SAV1, phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 inhibits its translocation into the nucleus to regulate cellular genes important for cell proliferation, cell death, and cell migration. Phosphorylates 'Ser-14' of histone H2B (H2BS14ph) during apoptosis. Phosphorylates FOXO3 upon oxidative stress, which results in its nuclear translocation and cell death initiation ().
