Intended Uses: This ALK1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep ALK1. This ELISA kit for research use only!

Principle of the Assay: ALK1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ALK1 antibody and an ALK1-HRP conjugate. The assay sample and buffer are incubated together with ALK1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ALK1 concentration since ALK1 from samples and ALK1-HRP conjugate compete for the anti-ALK1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ALK1 from the sample, fewer sites are left to bind ALK1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ALK1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a type I cell-surface receptor for the TGF-beta superfamily of ligands. It shares with other type I receptors a high degree of similarity in serine-threonine kinase subdomains, a glycine- and serine-rich region (called the GS domain) preceding the kinase domain, and a short C-terminal tail. The encoded protein, sometimes termed ALK1, shares similar domain structures with other closely related ALK or activin receptor-like kinase proteins that form a subfamily of receptor serine/threonine kinases. Mutations in this gene are associated with hemorrhagic telangiectasia type 2, also known as Rendu-Osler-Weber syndrome 2. [provided by RefSeq, Jul 2008]
