Intended Uses: This PRKACB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human PRKACB. This ELISA kit for research use only!

Principle of the Assay: PRKACB ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-PRKACB antibody and an PRKACB-HRP conjugate. The assay sample and buffer are incubated together with PRKACB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PRKACB concentration since PRKACB from samples and PRKACB-HRP conjugate compete for the anti-PRKACB antibody binding site. Since the number of sites is limited, as more sites are occupied by PRKACB from the sample, fewer sites are left to bind PRKACB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PRKACB concentration in each sample is interpolated from this standard curve.

NCBI Description

cAMP is a signaling molecule important for a variety of cellular functions. cAMP exerts its effects by activating the cAMP-dependent protein kinase, which transduces the signal through phosphorylation of different target proteins. The inactive kinase holoenzyme is a tetramer composed of two regulatory and two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of regulatory subunits bound to four cAMP and two free monomeric catalytic subunits. Four different regulatory subunits and three catalytic subunits have been identified in humans. The protein encoded by this gene is one of the regulatory subunits. This subunit can be phosphorylated by the activated catalytic subunit. It may interact with various A-kinase anchoring proteins and determine the subcellular localization of cAMP-dependent protein kinase. This subunit has been shown to regulate protein transport from endosomes to the Golgi apparatus and further to the endoplasmic reticulum (ER). [provided by RefSeq, Jul 2008]

PKAR2A: a regulatory subunit of cAMP-regulated protein kinase. The inactive form of the enzyme is composed of two regulatory chains and two catalytic chains. Activation by cAMP produces two active catalytic monomers and a regulatory dimer that binds four cAMP molecules. Four types of regulatory chains are found: I-alpha, I-beta, II-alpha, and II-beta. Their expression varies among tissues and is in some cases constitutive and in others inducible.
