Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Pro-HGF-A is a 96kD member of the PA-FXII-HGFA family of enzymes. It is secreted by hepatocytes and is found in plasma at ~40nM concentration. Other cells selectively reported to express HGF-A include astrocytes, endothelium, fibroblasts and macrophages. Upon activation, HGF-A is known to activate both proHGF and proMSP via proteolytic cleavage. The circulating 96kD form of mouse HGF-A represents an inactive 619aa proform aa35-653. Within this proform exists one FN type II domain aa100-147, an EGF-like domain aa157-195, an FN type III domain aa197-237, a second EGF-like domain aa238-276, a kringle domain aa283-364 and a functional peptidase S1 domain aa406-644. Activation of HGF-A typically occurs with a thrombin-mediated cleavage between aa405-406 and a KLK cleavage between aa369-370. This creates a 34kD heterodimeric active product that contains a 2kD N-terminus aa370-405 disulfide linked to a 32kD C-terminus aa406-653. Once activated, the 34kD heterodimer binds to cell surface HAI-1, rendering it unavailable to circulating HGF. When needed, this HGF-A:HAI-1 complex is released, HGF-A dissociates from HAI-1, and HGF-A is free to activate proHGF. Over aa35-653, mouse HGF-A shares 92% and 82% aa identity with rat and human HGF-A.

NCBI Description

This gene encodes a serine protease enzyme that proteolytically activates hepatocyte growth factor (HGF) and plays a vital role in the regulation of HGF activity in the regeneration and repair of various tissues. The encoded protein is an inactive zymogen that is proteolytically activated to generate a heterodimeric enzyme consisting of a short chain and a long chain linked by a disulfide bridge. Mice lacking the encoded protein display an impairment in mucosal regeneration after injury. [provided by RefSeq, Jul 2015]
