Western Blot (WB)
Application Notes
Suitable for use in Western Blot analysis and is not suitable for other applications. For Westerns, resuspend the protein in 20ul (1ug/ul) of a 1X SDS-PAGE sample buffer (6). On a 15-well minigel system, 100ng of protein per lane should be sufficient when used in a colorimetric Western Blot.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -70 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 12 months.

The 71kD antigen from M. tuberculosis is recognized by antibodies and by T lymphocytes during infection (1) and sequence analysis indicates a relationship between this antigen and the highly conserved Hsp70 family (2). Biochemical as well as serological characterization of the M. tuberculosis 71kD antigen confirms that it is a member of the Hsp70 family and metabolic labeling demonstrates that it is a major component of the mycobacterial response to heat stress. M. tuberculosis Hsp71 may have a biochemical role similar to that of DnaK which is involved in ATP-dependent protein-protein interactions and like eukaryotic Hsp70 homologs, is thought to participate in the assembly/disassembly of protein complexes (3, 4). Several studies have shown that during cellular rejection, the allograft undergoes a stress response which regulates the expression of stress proteins and triggers the recruitment and activation of hsp-reactive lymphocytes. Recent data using the rat model of heterotropic heart transplants, demonstrates that allograft-infiltrating lymphocytes respond to recombinant mycobacterial hsp71 and irradiated syngenic spleen cells as a source of self-APC. The hsp71-dependency of self-APC reactive T cell reactivity represents a previously unrecognized mechanism of cellular immunity to allografts. This mechanism might be related to the peptide binding properties of hsp71 and the ability of stress proteins to function as molecular chaperones in antigen processing (5).
