Specificity
6B10 reacts exlcusively with cytokeRatin 4 which is present in non-cornifying squamous epithelium, including cornea and transitional epithelium. Cells in certain ciliated pseudo-stRatified epithelia and ductal epithelia of various exocrine glands are also positive for 6B10.

Application Notes
6B10 is useful for immunocytochemistry, immunohistochemistry on frozen and tissues, immunoblotting and flow cytometry. Optimal antibody dilution should be determined by titRation; recommended range is 1:25 - 1:200 for flow cytometry, and for immunohistochemistry with avidin-biotinylated Horseradish peroxidase complex (ABC) as detection reagent, and 1:100 - 1:1000 for immunoblotting appliCations.

CytokeRatins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in Human epithelial tissues by at least 20 different polypeptides. They range in molecular weight between 40 kDa and 68 kDa and isoelectric pH between 4.9 - 7.8. The individual Human cytokeRatins are numbered 1 to 20. The various epithelia in the Human body usually express cytokeRatins which are not only characteristic of the type of epithelium, but also related to the degree of matuRation or differentiation within an epithelium. CytokeRatin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies. The cytokeRatin antibodies are not only of assistance in the differential identification of tumors using immunohistochemistry on tissue sections, but are also a useful tool in cytopathology and flow cytometric assays.

NCBI Description

The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in differentiated layers of the mucosal and esophageal epithelia with family member KRT13. Mutations in these genes have been associated with White Sponge Nevus, characterized by oral, esophageal, and anal leukoplakia. The type II cytokeratins are clustered in a region of chromosome 12q12-q13. [provided by RefSeq]

Uniprot Description

K4: Heterotetramer of two type I and two type II keratins. Keratin-4 is generally associated with keratin-13. Detected in the suprabasal layer of the stratified epithelium of the esophagus, exocervix, vagina, mouth and lingual mucosa, and in cells and cell clusters in the mucosa and serous gland ducts of the esophageal submucosa. Expressed widely in the exocervix and esophageal epithelium, with lowest levels detected in the basal cell layer. Belongs to the intermediate filament family.
