Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Liver X receptor alpha (LXR alpha), a NR1 Thyroid Hormone-Like Receptor, has been shown to affect the transport and catabolism of cholesterol as well as fatty acid metabolism. LXR alpha also appears to be an important insulin mediating factor in the regulation of lipogenesis. LXR alpha is known to be activated by oxysterols and to regulate a wide range of genes, including members of the ATP-binding cassette (ABC) superfamily of transporter proteins (ABCA1 and ABCG1), surface constituents of plasma lipoproteins like apolipoprotein E, and cholesterol ester transport protein. The induced expression of LXR alpha may be related to foam-cell formation in atherosclerotic lesions. As a result, LXR alpha has been suggested as a potential therapeutic target for treating atherosclerosis. LXR alpha forms a heterodimer with RXR alpha and binds the DR4-type cis-element. Recently LXR alpha has been shown to act as a cAMP-responsive nuclear modulator of renin and c-myc expression, suggesting a possible involvement of LXR alpha in the cardiovascular system and in general gene transcription.

NCBI Description

The protein encoded by this gene belongs to the NR1 subfamily of the nuclear receptor superfamily. The NR1 family members are key regulators of macrophage function, controlling transcriptional programs involved in lipid homeostasis and inflammation. This protein is highly expressed in visceral organs, including liver, kidney and intestine. It forms a heterodimer with retinoid X receptor (RXR), and regulates expression of target genes containing retinoid response elements. Studies in mice lacking this gene suggest that it may play an important role in the regulation of cholesterol homeostasis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]

Interaction with RXR shifts RXR from its role as a silent DNA-binding partner to an active ligand- binding subunit in mediating retinoid responses through target genes defined by LXRES. LXRES are DR4-type response elements characterized by direct repeats of two similar hexanuclotide half- sites spaced by four nucleotides. Plays an important role in the regulation of cholesterol homeostasis, regulating cholesterol uptake through MYLIP-dependent ubiquitination of LDLR, VLDLR and LRP8. Belongs to the nuclear hormone receptor family. NR1 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
