Description: The antibody is a rabbit polyclonal antibody made to the serine proteinase kallikrein-15 (hK15). The antibody is made to a synthetic peptide based on the catalytic domain of human kallikrein-15. The antibody reacts with human kallikrein-15, and does not react with the human kallikreins (hK1-14). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. Kallikrein-15, also known as prostinogen and HSRNASPH, is a trypsin-like serine proteinase. Kallikrein-15 was discovered as a novel serine protease in brain tissue from Alzheimer's patients. Later Kallikrein-15 was cloned from prostate tissue, and named prostinogen. The zymogen of Kallikrein-15 was termed prostinogen, and the active form is termed prostin.Independently discovered by cloning the kallikrein locus on chromosome 19, Kallikrein-15 was identified by homology to the other kallikrein family genes. Kallikrein-15 is produced by a number of tissues, including the brain, spinal cord, liver, salivary gland, adrenal gland, prostate, colon, thymus and thyroid. Like many of the kallikreins, kallikrein-15 is also found in biological fluids like serum, breast milk, amniotic fluid, seminal plasma and cerebrospinal fluid. Kallikrein-15 message has been reported to be decreased in some breast cancers, but increased in ovarian cancers and prostate cancer. Some cell lines express kallikrein-15 in culture, and like many of the kallikreins, the production is stimulated by hormones. Kallikrein-15 was found to activate kallikrein-3 (PSA) in vitro, and prostate tumors producing both kallikrein-3 and Kallikrein-15 were found to be more aggressive Endogenous inhibitors include kallistatin, protein-C inhibitor and beta1-proteinase inhibitor, although Kallikrein-15 can be found complexed to a number of different proteinase inhibitors. A number of slice variants of Kallikrein-15 have been discovered. The longest kallikrein-15 sequence codes for a protein of 256 amino acids, with predicted mass of 28.1 kDa, and a pI of 9.3. The 171 amino acid form is predicted at 19.3 kDa and 9.8 pI, the 162 amino acid form at 18 kDa and 11.3 pI, a 161 amino acid form at 17.9 kDa and 11.3 pI and the 121 amino acid form 13.9 kDa and 10.9 pI. It is not clear how many of these sequences are produced as proteins. Glycosylation and other post-translational modifications make Kallikrein-15 run at a higher molecular weight on SDS PAGE gels. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species.

Relevance: The human tissue Kallikrein gene family encodes 15 serine proteases. All Kallikreins share structural similarities including cysteine residues, a catalytic triad of His, Asp, and Ser residues, typically five coding exons, and varied intron phases. Kallikreins are predominantly secreted as inactive zymogens prior to activation by cleavage of an N-terminal peptide, and all function extracellularly. Kallikreins can be activated autocatalytically, via other Kallikreins, or additional proteases. While structurally similar, Kallikrein family members have distinct functions and have key roles in many physiological and pathological processes. Many human tissue Kallikreins also show promise as cancer biomarkers, which may facilitate earlier detection and characterization of many forms of cancer. Kallikrein 15 is one of the fifteen kallikrein subfamily members whose gene is located in a cluster on chromosome 19. Increased levels are found in prostate cancer, which indicates its possible use as a prognostic marker for prostate cancer. Four splice variants, each encoding a distinct isoform, have been described.

NCBI Description

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. In prostate cancer, this gene has increased expression, which indicates its possible use as a diagnostic or prognostic marker for prostate cancer. The gene contains multiple polyadenylation sites and alternative splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Jul 2008]
