Application Notes
WB: Use at an assay dependent dilution. Predicted molecular weight: 27 kDa. A recommended starting concentration for Western blots is 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species. A range of different sized bands can be seen for Kallikrein 6 in different tissues and in cell culture media, ranging from 16-32 kDa and likely representing different proteolytically processed forms, but perhaps also the different splice variants. Dilution optimised using Chromogenic detection.

Description: The antibody is a rabbit polyclonal antibody made to the serine proteinase kallikrein-6 (hK6). The antibody is made to a synthetic peptide based on the Kallikrein loop area of human Kallikrein-6. The antibody reacts with human Kallikrein-6, and does not react with the human kallikreins (hK1-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. Kallikrein-6, also known as Zyme, Neurosin, PRSS9, myelencephalon-specific protease (MSP) and protease M, is a trypsin-like serine proteinase. Kallikrein-6 was originally described from the brain as an enzyme involved in degradation of amyloid plaque protein (APP), and hK6 was thought to be a beta secretase. Kallikrein-6 was shown to be elevated in the sera patients with Alzheimer's disease and Parkinson's disease, and in animal models of multiple sclerosis. The neuronal protein alpha-synuclein was shown to be cleaved by hK6, as has APP, and hK6 has been implicated in vascular morphogenesis and angiogenesis. MSP has also been shown to rapidly degrade myelin-specific protein, laminin and fibronectin. Kallikrein-6 is found is greatest abundance in the brain and neuronal tissues, with lesser levels in the kidney, thymus, spleen and lymph nodes. Human breast milk is reported to contain high levels of Kallikrein-6, and hK6 has bee purified from cerebral spinal fluid (CSF). Less stringent PCR detects hK6 in many tissues, and analysis of cell lines by Western blot shows that many cell lines make hK6. Endogenous inhibitors include kallistatin, protein-C inhibitor and beta1-proteinase inhibitor, although Kallikrein-6 can be found complexed to a number of different proteinase inhibitors. Pre-pro-kallikrein-6 has the 16 amino acid signal sequence that is removed before secretion, and the Pro-kallikrein-5 is activated to Kallikrein-5 by removal of the 5 amino acid propeptide domain. The full length hK6 sequence codes for a protein of 244 amino acids, with predicted mass of 26.9 kDa, and a pI of 7.53, and the processed hK6 has an apparent mass of 31-33 kDa on reduced SDS PAGE gels. NCBI Description

This gene encodes a member of the kallikrein subfamily of the peptidase S1 family of serine proteases. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. The encoded preproprotein is proteolytically processed to generate the mature protease. Expression of this protease is regulated by steroid hormones and may be elevated in multiple human cancers and in serum from psoriasis patients. The encoded protease may participate in the cleavage of amyloid precursor protein and alpha-synuclein, thus implicating this protease in Alzheimer's and Parkinson's disease, respectively. This gene is located in a gene cluster on chromosome 19. Alternative splicing results in multiple transcript variants, at least one of which encodes an isoform that is proteolytically processed. KLK6: Serine protease which exhibits a preference for Arg over Lys in the substrate P1 position and for Ser or Pro in the P2 position. Shows activity against amyloid precursor protein, myelin basic protein, gelatin, casein and extracellular matrix proteins such as fibronectin, laminin, vitronectin and collagen. Degrades alpha-synuclein and prevents its polymerization, indicating that it may be involved in the pathogenesis of Parkinson disease and other synucleinopathies. May be involved in regulation of axon outgrowth following spinal cord injury. Tumor cells treated with a neutralizing KLK6 antibody migrate less than control cells, suggesting a role in invasion and metastasis. Belongs to the peptidase S1 family. Kallikrein subfamily. 3 isoforms of the human protein are produced by alternative splicing.
