Principle of the assay: XPC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-XPC antibody and an XPC-HRP conjugate. The assay sample and buffer are incubated together with XPC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the XPC concentration since XPC from samples and XPC-HRP conjugate compete for the anti-XPC antibody binding site. Since the number of sites is limited, as more sites are occupied by XPC from the sample, fewer sites are left to bind XPC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The XPC concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a component of the nucleotide excision repair (NER) pathway. There are multiple components involved in the NER pathway, including Xeroderma pigmentosum (XP) A-G and V, Cockayne syndrome (CS) A and B, and trichothiodystrophy (TTD) group A, etc. This component, XPC, plays an important role in the early steps of global genome NER, especially in damage recognition, open complex formation, and repair protein complex formation. Mutations in this gene or some other NER components result in Xeroderma pigmentosum, a rare autosomal recessive disorder characterized by increased sensitivity to sunlight with the development of carcinomas at an early age. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Mar 2009]
