Scientific Background: VRK3 is a member of the vaccinia-related kinase (VRK) family of serine/threonine protein kinases which has substitutions at several residues within the ATP binding motifs that in other kinases have been shown to be required for catalysis. Profound changes to the active site region underlie the loss of catalytic activity of VRK3 since it cannot bind ATP because of residue substitutions in the binding pocket (1). VRK3 is widely expressed in human tissues and the protein localizes to the nucleus. VRK3 regulates several transcription factors, nuclear envelope assembly, and chromatin condensation and is also required for cell cycle progression (2).

NCBI Description

This gene encodes a member of the vaccinia-related kinase (VRK) family of serine/threonine protein kinases. In both human and mouse, this gene has substitutions at several residues within the ATP binding motifs that in other kinases have been shown to be required for catalysis. In vitro assays indicate the protein lacks phosphorylation activity. The protein, however, likely retains its substrate binding capability. This gene is widely expressed in human tissues and its protein localizes to the nucleus. Alternative splicing results in multiple transcripts encoding different isoforms. Uniprot Description

Function: Inactive kinase that suppresses ERK activity by promoting phosphatase activity of DUSP3 which specifically dephosphorylates and inactivates ERK in the nucleus

By similarity. Ref.6 Ref.12

Subunit structure: Interacts with DUSP3

By similarity. Interacts with RAN. Ref.7

Subcellular location: Nucleus Ref.6.

Sequence similarities: Belongs to the protein kinase superfamily. CK1 Ser/Thr protein kinase family. VRK subfamily.Contains 1 protein kinase domain.

Caution: Inactive as a kinase due to its inability to bind ATP (Ref.12).

Product Notes

The VRK3 vrk3 (Catalog #AAA515272) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant full-length human VRK3 was expressed by baculovirus in Sf9 insect cells. AAA Biotech's VRK3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the VRK3 vrk3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "VRK3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
