Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Voltage Dependent Anion Channel Protein 1 (VDAC1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Voltage Dependent Anion Channel Protein 1 (VDAC1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Voltage Dependent Anion Channel Protein 1 (VDAC1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Voltage Dependent Anion Channel Protein 1 (VDAC1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

Encodes a voltage-dependent anion channel (VDAC: AT3G01280/VDAC1, AT5G67500/VDAC2, AT5G15090/VDAC3, AT5G57490/VDAC4, AT5G15090/VDAC5). VDACs are reported to be porin-type, beta-barrel diffusion pores. They are prominently localized in the outer mitochondrial membrane and are involved in metabolite exchange between the organelle and the cytosol.

Uniprot Description

Forms a channel through the mitochondrial outer membrane that allows diffusion of small hydrophilic molecules. The channel adopts an open conformation at low or zero membrane potential and a closed conformation at potentials above 30-40 mV. The open state has a weak anion selectivity whereas the closed state is cation-selective (). Involved in plant development at reproductive stage, is important for pollen development and may regulate hydrogen peroxide generation during disease resistance.
