Principle of the Assay||The Mouse GITRL ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse GITRL in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse GITRL coated on a 96-well plate. Standards and samples are pipetted into the wells and GITRL present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse GITRL antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of GITRL bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Tumor necrosis factor ligand superfamily member 18, also known as AITRL or GITRL, is a protein that in humans is encoded by the TNFSF18 gene. The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor(TNF) ligand family. It is mapped to 1q25.1. TNFSF18 is a ligand for receptor TNFRSF18/AITR/GITR. It has been shown to modulate T lymphocyte survival in peripheral tissues. This cytokine is also found to be expressed in endothelial cells, and is thought to be important for interaction between T lymphocytes and endothelial cells. TNFSF18-dependent modulation of tryptophan catabolism may represent an important mechanism of action of glucocorticoids, both physiologically and therapeutically.

Uniprot Description

TNFSF18: Cytokine that binds to TNFRSF18/AITR/GITR. Regulates T- cell responses. Can function as costimulator and lower the threshold for T-cell activation and T-cell proliferation. Important for interactions between activated T-lymphocytes and endothelial cells. Mediates activation of NF-kappa-B. Belongs to the tumor necrosis factor family.

Protein type: Membrane protein, integral; Cytokine; Apoptosis

Cellular Component: cell surface; extracellular space; integral to membrane; membrane; plasma membrane

Molecular Function: cytokine activity; protein binding; tumor necrosis factor receptor binding; tumor necrosis factor receptor superfamily binding

Product Notes

The Mouse TNFSF18 tnfsf18 (Catalog #AAA8244685) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244685 ELISA Kit recognizes Mouse TNFSF18. AAA Biotech's GITRL can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the TNFSF18 tnfsf18 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GITRL, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
