Related Product Information for TNFSF14/LIGHT/CD258 recombinant protein
Recombinant Human TNFSF14/LIGHT/CD258 Protein is produced by HEK293 cells expression system. The target protein is expressed with sequence (Asp74-Val240) of Human TNFSF14 (Accession #NP_003798.2) fused with a 6×His tag at the N-terminus.
Background: LIGHT, also known as TNFSF14 or CD258, is a newly identified member of the TNF superfamily (TNFSF14) that is expressed by activated T lymphocytes, monocytes, granulocytes, spleen cells, and immature dendritic cells. TNFSF14/LIGHT/CD258 is a type II transmembrane protein that is known to bind 2 membrane-bound TNFSF signaling receptors: HVEM, which is predominantly expressed by T cells, and lymphotoxin beta receptor (LTbetaR), which is expressed by stromal cells and nonlymphoid hematopoietic cells. TNFSF14/LIGHT/CD258 also binds to a soluble non-signaling receptor, decoy receptor 3 (DcR3), which can modulate the function of LIGHT in vivo. TNFSF14/LIGHT/CD258 can also costimulate T cell responses via HVEM, which is constitutively expressed in most lymphocyte subpopulations, including CD4+and CD8+T cells. In addition, TNFSF14/LIGHT/CD258 has been shown to suppress tumor formation in vivo and to induce tumor cell apoptosis via the up-regulation of intercellular adhesion molecule 1 and an increased lymphocyte adhesion to cancer cells. Thus, TNFSF14/LIGHT/CD258 is being actively investigated as a possible basis for cancer treatment.

NCBI Description

The protein encoded by this gene is a member of the tumor necrosis factor (TNF) ligand family. This protein is a ligand for TNFRSF14, which is a member of the tumor necrosis factor receptor superfamily, and which is also known as a herpesvirus entry mediator (HVEM). This protein may function as a costimulatory factor for the activation of lymphoid cells and as a deterrent to infection by herpesvirus. This protein has been shown to stimulate the proliferation of T cells, and trigger apoptosis of various tumor cells. This protein is also reported to prevent tumor necrosis factor alpha mediated apoptosis in primary hepatocyte. Two alternatively spliced transcript variant encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

Uniprot Description

TNFSF14: Cytokine that binds to TNFRSF3/LTBR. Binding to the decoy receptor TNFRSF6B modulates its effects. Activates NFKB, stimulates the proliferation of T-cells, and inhibits growth of the adenocarcinoma HT-29. Acts as a receptor for Herpes simplex virus. Belongs to the tumor necrosis factor family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Cytokine; Inhibitor

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: extracellular space; cytoplasm; plasma membrane; integral to membrane

Molecular Function: protein binding; caspase inhibitor activity; cytokine activity; tumor necrosis factor receptor binding; receptor binding

Biological Process: release of cytoplasmic sequestered NF-kappaB; T cell proliferation; T cell activation; apoptosis; T cell costimulation; T cell homeostasis; immune response; negative regulation of caspase activity; signal transduction; positive regulation of myoblast differentiation

Product Notes

The TNFSF14/LIGHT/CD258 tnfsf14 (Catalog #AAA9144215) is a Recombinant Protein produced from HEK293 cells and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "TNFSF14/LIGHT/CD258, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
