Related Product Information for TNFRSF11A elisa kit
Principle of the Assay: The Picokine™ Mouse Tnfrsf11a Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse Tnfrsf11a with a 96-well strip plate that is pre-coated with antibody specific for Tnfrsf11a. The detection antibody is a biotinylated antibody specific for Tnfrsf11a. The capture antibody is monoclonal antibody from rat, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Mouse Tnfrsf11a with immunogen: Expression system for standard: NSO; Immunogen sequence: Q30-P213. The kit is analytically validated with ready to use reagents. To measure Mouse Tnfrsf11a, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Mouse Tnfrsf11a in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Tnfrsf11a in the sample.

Background/Introduction: Receptor Activator of Nuclear Factor kappa B(RANK), also known as TRANCE Receptor, is a type I membrane protein that is expressed on the surface of osteoclasts and is involved in their activation upon ligand binding. RANK is a recently described TNF receptor family member, and its ligand, RANKL, promote survival of dendritic cells and differentiation of osteoclasts. RANK contains 383 amino acids in its intracellular domain(residues 234-616), which contain three putative TRAF-binding domains(termed I, II, and III). RANK interacts with various TRAFs through distinct motifs and activates NF-kappaB via a novel TRAF6 interaction motif, which then activates NIK, thus leading to NF-kappaB activation, whereas RANK most likely activates JNK through a TRAF2-interacting region in RANK.

Uniprot Description

TNFRSF11A: Receptor for TNFSF11/RANKL/TRANCE/OPGL; essential for RANKL-mediated osteoclastogenesis. Involved in the regulation of interactions between T-cells and dendritic cells. Binds to the clefts between the subunits of the TNFSF11 ligand trimer to form a heterohexamer. Interacts with TRAF1, TRAF2, TRAF3, TRAF5 and TRAF6. Interacts (via cytoplasmic domain) with GAB2. Ubiquitous expression with high levels in skeletal muscle, thymus, liver, colon, small intestine and adrenal gland.

Product Notes

The Mouse TNFRSF11A tnfrsf11a (Catalog #AAA175876) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA175876 ELISA Kit recognizes Mouse TNFRSF11A. It is sometimes possible for the material contained within the vial of "RANK, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
