Principle of the Assay: The Human Tenascin C ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Tenascin C in Cell Culture Supernates, Serum, Plasma, Urine. This assay employs an antibody specific for Human Tenascin C coated on a 96-well plate. Standards and samples are pipetted into the wells and Tenascin C present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Tenascin C antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Tenascin C bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background: Tenascin C (TN-C) is a glycoprotein that in humans is encoded by the TNC gene. It is expressed in the extracellular matrix of various tissues during development, disease or injury, and in restricted neurogenic areas of the central nervous system. Tenascin-C is the founding member of the gene family (Tenascin). In the embryo it is made by migrating cells like the neural crest; it is also abundant in developing tendons, bone and cartilage. TN-C clearly plays a role in cell signaling as evidenced by its ability to be induced during events such as trauma, inflammation, or cancer development. Also, TN-C is important in regulating cell proliferation and migration, especially during developmental differentiation and wound healing.
NCBI Description
This gene encodes an extracellular matrix protein with a spatially and temporally restricted tissue distribution. This protein is homohexameric with disulfide-linked subunits, and contains multiple EGF-like and fibronectin type-III domains. It is implicated in guidance of migrating neurons as well as axons during development, synaptic plasticity, and neuronal regeneration. [provided by RefSeq, Jul 2011]
Uniprot Description
TNC: Extracellular matrix protein implicated in guidance of migrating neurons as well as axons during development, synaptic plasticity as well as neuronal regeneration. Promotes neurite outgrowth from cortical neurons grown on a monolayer of astrocytes. Ligand for integrins alpha-8/beta-1, alpha-9/beta-1, alpha-V/beta-3 and alpha-V/beta-6. Belongs to the tenascin family. 6 isoforms of the human protein are produced by alternative splicing.

The Human TNC tnc (Catalog #
AAA8248408) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8248408 ELISA Kit recognizes Human TNC.
It is sometimes possible for the material contained within the vial of
"Tenascin C, ELISA Kit" to become dispersed throughout the inside of
the vial, particularly around the seal of said vial, during shipment and storage. We always
suggest centrifuging these vials
to consolidate all of the liquid away from the lid and to the bottom of the vial prior to
opening. Please be advised that
certain products may require dry ice for shipping and that, if this is the case, an
additional dry ice fee may also be
required.
