Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of mouse CIT gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

Uniprot Description

CRIK: an AGC kinase that is a RHO/RAC effector. Binds to the GTP-bound forms of RHO and RAC1. Required for postnatal hippocampal neurogenesis. Rho/CRIK suppresses differentiation and is associated with induction of KyoT1/2, a LIM domain protein gene implicated in integrin-mediated processes and Notch signaling. Plays an important role in the regulation of cytokinesis and the development of the central nervous system. Overexpressed in mantle-cell lymphoma (MCL), an aggressive type of B-cell lymphoma. Three alternatively spliced isoforms have been described.

Protein type: EC 2.7.11.1; Protein kinase, Ser/Thr (non-receptor); Kinase, protein; Protein kinase, AGC; AGC group; DMPK family

Cellular Component: ruffle; Golgi cisterna; membrane; cell soma; cytoplasm; plasma membrane; vacuole; actin cytoskeleton

Molecular Function: transferase activity; protein serine/threonine kinase activity; protein binding; Rho GTPase binding; metal ion binding; transferase activity, transferring phosphorus-containing groups; nucleotide binding; kinase activity; SH3 domain binding; ATP binding; PDZ domain binding; protein kinase activity

Product Notes

The CIT cit (Catalog #AAA8216094) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The CIT siRNA (Mouse) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CIT can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the CIT cit for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CIT, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
