Principle of the Assay||The microtiter plate provided in this kit has been pre-coated with an antibody specific to Solute carrier family 40 member 1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Solute carrier family 40 member 1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Solute carrier family 40 member 1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Solute carrier family 40 member 1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

SLC40A1: May be involved in iron export from duodenal epithelial cell and also in transfer of iron between maternal and fetal circulation. Mediates iron efflux in the presence of a ferroxidase (hephaestin and/or ceruloplasmin). Defects in SLC40A1 are the cause of hemochromatosis type 4 (HFE4). HFE4 is an autosomal dominant iron-loading disorder characterized by early iron accumulation in reticuloendothelial cells and a marked increase in serum ferritin before elevation of the transferrin saturation. Belongs to the ferroportin (FP) (TC 2.A.100) family. SLC40A subfamily.
