Principle of the Assay: SLC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SLC antibody and an SLC-HRP conjugate. The assay sample and buffer are incubated together with SLC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SLC concentration since SLC from samples and SLC-HRP conjugate compete for the anti-SLC antibody binding site. Since the number of sites is limited, as more sites are occupied by SLC from the sample, fewer sites are left to bind SLC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SLC concentration in each sample is interpolated from this standard curve.

NCBI Description

This antimicrobial gene is one of several CC cytokine genes clustered on the p-arm of chromosome 9. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. Similar to other chemokines the protein encoded by this gene inhibits hemopoiesis and stimulates chemotaxis. This protein is chemotactic in vitro for thymocytes and activated T cells, but not for B cells, macrophages, or neutrophils. The cytokine encoded by this gene may also play a role in mediating homing of lymphocytes to secondary lymphoid organs. It is a high affinity functional ligand for chemokine receptor 7 that is expressed on T and B lymphocytes and a known receptor for another member of the cytokine family (small inducible cytokine A19).
