Biological Significance
14-3-3 sigma, also known as Stratifin (SFN), belongs to the 14-3-3 family. The 14-3-3 family of proteins plays a key regulatory role in signal transduction, checkpoint control, apoptotic and nutrient-sensing pathways. 14-3-3 proteins are highly conserved and ubiquitously expressed. There are at least seven isoforms, beta, gamma, epsilon, sigma, zeta, tau and eta that have been identified in mammals. 14-3-3 sigma was identified as an epithelial cell marker and appeared to function as a tumor suppressor whose expression can be down regulated via methylation. Loss of 14-3-3 sigma expression results in a defective G2/M phase checkpoint and appears to contribute to both epithelial and non-epithelial tumorigenesis. Recombinant human 14-3-3 sigma was expressed in E Coli and purified by using conventional chromatography techniques.
Expression System
E Coli
Preparation and Storage
Store at 4 degree C for short term storage (1/2 weeks). Aliquot and store at -20 degree C or - 70 degree C for long term storage. Avoid repeated freeze/thaw cycles.

Uniprot Description

14-3-3 sigma: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. When bound to KRT17, regulates protein synthesis and epithelial cell growth by stimulating Akt/mTOR pathway. Homodimer. Interacts with KRT17 and SAMSN1. Found in a complex with XPO7, EIF4A1, ARHGAP1, VPS26A, VPS29, VPS35 and SFN. Interacts with GAB2. Interacts with SRPK2. Present mainly in tissues enriched in stratified squamous keratinizing epithelium. Belongs to the 14-3-3 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The 14.3.3 sigma sfn (Catalog #AAA836352) is a Recombinant Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's 14.3.3 sigma can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Researchers should empirically determine the suitability of the 14.3.3 sigma sfn for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: 1-248 amino acids: MERASLIQKA KLAEQAERYE DMAAFMKGAV EKGEELSCEE RNLLSVAYKN VVGGQRAAWR VLSSIEQKSN EEGSEEKGPE VREYREKVET ELQGVCDTVL GLLDSHLIKE AGDAESRVFY LKMKGDYYRY LAEVATGDDK KRIIDSARSA YQEAMDISKK EMPPTNPIRL GLALNFSVFH YEIANSPEEA ISLAKTTFDE AMADLHTLSE DSYKDSTLIM QLLRDNLTLW TADNAGEEGG EAPQEPQS. It is sometimes possible for the material contained within the vial of "14.3.3 sigma, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
