* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: Thyrotropin-releasing hormone (TRH) acts as a stimulator of hormone secretion from adenohypophyseal cells, and its signals are inactivated by TRH-degrading ectoenzyme. The deduced 1,024-amino acid type II integral transmembrane protein is 96% identical to the rat protein. It has 12 putative C-terminal N-glycosylation sites, a potential tyrosine sulfation site (residue 380), a consensus sequence for zinc-dependent metallopeptidases, and a short N-terminal intracellular domain with a potential phosphorylation site. Northern blot analysis revealed expression of 7.5-, 7.0-, 6.0-, and 4.5-kb transcripts. Strongest expression was in brain, with weaker expression in heart, lung, liver, and skeletal muscle, and no expression in kidney and placenta.

This gene encodes a member of the thyrotropin-releasing hormone family. Cleavage of the encoded proprotein releases mature thyrotropin-releasing hormone, which is a tripeptide hypothalamic regulatory hormone. The rat proprotein contains five thyrotropin-releasing hormone tripeptides. Thyrotropin-releasing hormone is involved in the regulation and release of thyroid-stimulating hormone, as well as prolactin, and may be involved in cardiac function in heart failure. Disruption of this gene is associated with hypothyroidism, elevated thyroid-stimulating hormone levels, and hyperglycemia. [provided by RefSeq, May 2013]
