Rat Signal Transducer And Activator Of Transcription 3 (STAT3) ELISA Kit | STAT3 elisa kit

Rat Signal Transducer And Activator Of Transcription 3 (STAT3) ELISA Kit

Reactivity
Rat
Synonyms
Signal Transducer And Activator Of Transcription 3 (STAT3); Rat Signal Transducer And Activator Of Transcription 3 (STAT3) ELISA Kit; STAT3 elisa kit
Ordering
For Research Use Only!
Reactivity
Rat
Specificity
This assay has high sensitivity and excellent specificity for detection of STAT3. No significant cross-reactivity or interference between STAT3 and analogues was observed.
Sequence Length
770
Samples
Rat tissue homogenates and other biological fluids
Detection Range
0.156-10ng/mL
Sensitivity
The minimum detectable dose of rat STAT3 is typically less than 0.054ng/mL. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined by adding two standard deviations to the mean optical density value of twenty zero standard replicates and calculating the corresponding concentration.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level STAT3 were tested 20 times on one plate, respectively. Intra-Assay: CV<10%
Inter-assay Precision
Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level STAT3 were tested on 3 different plates, 8 replicates in each plate. CV(%) = SD/meanX100. Inter-Assay: CV<12%
Intended Uses
The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of STAT3 in rat tissue homogenates and other biological fluids.
Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Principle of the assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to STAT3. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to STAT3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain STAT3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of STAT3 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

Function: Signal transducer and transcription activator that mediates cellular responses to interleukins, KITLG/SCF and other growth factors. May mediate cellular responses to activated FGFR1, FGFR2, FGFR3 and FGFR4. Binds to the interleukin-6 (IL-6)-responsive elements identified in the promoters of various acute-phase protein genes. Activated by IL31 through IL31RA

By similarity. Cytoplasmic STAT3 represses macroautophagy by inhibiting EIF2AK2/PKR activity

By similarity. Plays an important role in host defense in methicillin-resistant S.aureus lung infection by regulating the expression of the antimicrobial lectin REG3G

By similarity.

Subunit structure: Forms a homodimer or a heterodimer with a related family member (at least STAT1). Interacts with IL31RA, NCOA1, PELP1, SIPAR, SOCS7, STATIP1 and TMF1. Interacts with IL23R in presence of IL23. Interacts (via SH2 domain) with NLK. Interacts with KPNA4 and KPNA5; KPNA4 may be the primary mediator of nuclear import. Interacts with CAV2; the interaction is increased on insulin-induced tyrosine phosphorylation of CAV2 and leads to STAT3 activation. Interacts with ARL2BP; interaction is enhanced with ARL2. Interacts with ARL2BP; the interaction is enhanced by LIF and JAK1 expression. Interacts with NEK6. Binds to CDK9 when activated and nuclear. Interacts with BMX. Interacts with EGFR; mediates EGFR downstream signaling to control cell proliferation. Interacts with STMN3, antagonizing its microtubule-destabilizing activity

By similarity. Interacts with the 'Lys-129' acetylated form of BIRC5/survivin. Interacts with FER

By similarity. Interacts with NEK6

By similarity. Interacts with BMX

By similarity. Interacts with ZIPK/DAPK3

By similarity. Interacts (via SH2 domain) with EIF2AK2/PKR (via the kinase catalytic domain)

By similarity.

Subcellular location: Cytoplasm. Nucleus. Note: Shuttles between the nucleus and the cytoplasm. Translocated into the nucleus upon tyrosine phosphorylation and dimerization, in response to signaling by activated FGFR1, FGFR2, FGFR3 or FGFR4. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly present in the cytoplasm without stimuli. Upon leukemia inhibitory factor (LIF) stimulation, accumulates in the nucleus. The complex composed of BART and ARL2 plays an important role in the nuclear translocation and retention of STAT3

By similarity. Ref.1

Tissue specificity: Detected in lung, heart, oviduct, ovary, uterus and kidney (at protein level). Detected in ovary, oviduct, and at lower levels in uterus and lung. Ref.1

Post-translational modification: Activated through tyrosine phosphorylation by BMX. Tyrosine phosphorylated in response to IL6, IL11, CNTF, LIF, KITLG/SCF, CSF1, EGF, PDGF, IFN-alpha and OSM. Activated KIT promotes phosphorylation on tyrosine residues and subsequent translocation to the nucleus. Tyrosine phosphorylated in response to constitutively activated FGFR1, FGFR2, FGFR3 and FGFR4. Phosphorylated on serine upon DNA damage, probably by ATM or ATR. Serine phosphorylation is important for the formation of stable DNA-binding STAT3 homodimers and maximal transcriptional activity. ARL2BP may participate in keeping the phosphorylated state of STAT3 within the nucleus. Tyrosine phosphorylated upon stimulation with EGF. Upon LPS challenge, phosphorylated within the nucleus by IRAK1. Phosphorylated on Ser-727 by RPS6KA5

By similarity. Phosphoryation at Tyr-705 by FER or PTK6 leads to an increase of its transcriptional activity

By similarity. Dephosphorylation on tyrosine residues by PTPN2 negatively regulates IL6/interleukin-6 signaling

By similarity. Ref.1

Miscellaneous: Involved in the gp130-mediated signaling pathway.

Sequence similarities: Belongs to the transcription factor STAT family.Contains 1 SH2 domain.

Product Notes

The Rat STAT3 stat3 (Catalog #AAA2019068) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2019068 ELISA Kit recognizes Rat STAT3. It is sometimes possible for the material contained within the vial of "Signal Transducer And Activator Of Transcription 3 (STAT3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
