* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: Galectin-9 (Gal-9) is a member of animal lectins that have an affinity to beta-galactosides. Gal-9 was first cloned from tumor tissues of Hodgkin's disease that is frequently characterized by eosinophilia. Galectins exhibit various biological functions, such as cell aggregation and adhesion, proliferation,cell death, modulation of inflammation and metastasis.First, Gal-9/ecalectin was identified as a potent T cellderived eosinophil chemoattractant (ECA). Gal-9 exhibits ECA activity in vitro and in vivo, activates eosinophils, and influences eosinophil survival. It requires divalent galactoside- binding activity but not its linker peptide to exhibit its functions, and some matrix metalloproteinases are involved in the release.

Astrocyte derived galectin-9 enhances microglial TNF production (PubMed:25158758). May play a role in thymocyte-epithelial interactions relevant to the biology of the thymus. May provide the molecular basis for urate flux across cell membranes, allowing urate that is formed during purine metabolism to efflux from cells and serving as an electrogenic transporter that plays an important role in renal and gastrointestinal urate excretion (PubMed:8995305). Highly selective to the anion urate (PubMed:8995305).
