Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Galectin-9 (LGALS9) ELISA Kit employs a two-site sandwich ELISA to quantitate LGALS9 in samples. An antibody specific for LGALS9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LGALS9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LGALS9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LGALS9 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Galectin-9 (Gal-9) is a member of animal lectins that have an affinity to beta-galactosides. Gal-9 was first cloned from tumor tissues of Hodgkin's disease that is frequently characterized by eosinophilia. Galectins exhibit various biological functions, such as cell aggregation and adhesion, proliferation,cell death, modulation of inflammation and metastasis.First, Gal-9/ecalectin was identified as a potent T cellderived eosinophil chemoattractant (ECA). Gal-9 exhibits ECA activity in vitro and in vivo, activates eosinophils, and influences eosinophil survival. It requires divalent galactoside- binding activity but not its linker peptide to exhibit its functions, and some matrix metalloproteinases are involved in the release.

NCBI Description

The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. The protein encoded by this gene is an S-type lectin. It is overexpressed in Hodgkin's disease tissue and might participate in the interaction between the H&RS cells with their surrounding cells and might thus play a role in the pathogenesis of this disease and/or its associated immunodeficiency. Multiple alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Jul 2008]
