Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for EGF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EGF present is bound by the immobilized antibody. Following incubation unbound samples are removed during a wash step, and then a detection antibody specific for EGF is added to the wells and binds to the combination of capture antibody-EGF in sample. Following a wash to remove any unbound combination, and enzyme conjugate is added to the wells. Following incubation and wash steps, a substrate is added. A colored product TMB is formed in proportion to the amount of EGF present in the sample. The reaction is terminated by addition of acid and absorbance is measured. A standard curve is prepared from seven EGF standard dilutions and EGF sample concentration determined.

Background/Introduction: EGF (Epidermal Growth Factor) is a Protein Coding gene. Diseases associated with EGF include Hypomagnesemia 4, Renal and Familial Primary Hypomagnesemia With Normocalciuria And Normocalcemia. Among its related pathways are PI-3K cascade-FGFR3 and p70S6K Signaling. GO annotations related to this gene include calcium ion binding and epidermal growth factor receptor binding. An important paralog of this gene is LRP1. This gene encodes a member of the epidermal growth factor superfamily. The encoded preproprotein is proteolytically processed to generate the 53-amino acid epidermal growth factor peptide. This protein acts a potent mitogenic factor that plays an important role in the growth, proliferation and differentiation of numerous cell types. This protein acts by binding with high affinity to the cell surface receptor, epidermal growth factor receptor. Defects in this gene are the cause of hypomagnesemia type 4. Dysregulation of this gene has been associated with the growth and progression of certain cancers. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed. EGF stimulates the growth of various epidermal and epithelial tissues in vivo and in vitro and of some fibroblasts in cell culture. Magnesiotropic hormone that stimulates magnesium reabsorption in the renal distal convoluted tubule via engagement of EGFR and activation of the magnesium channel TRPM6. Can induce neurite outgrowth in motoneurons of the pond snail Lymnaea stagnalis in vitro (PubMed:10964941).
