Principle of the assay: ADORA1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADORA1 antibody and an ADORA1-HRP conjugate. The assay sample and buffer are incubated together with ADORA1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADORA1 concentration since ADORA1 from samples and ADORA1-HRP conjugate compete for the anti-ADORA1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADORA1 from the sample, fewer sites are left to bind ADORA1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADORA1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is an adenosine receptor that belongs to the G-protein coupled receptor 1 family. There are 3 types of adenosine receptors, each with a specific pattern of ligand binding and tissue distribution, and together they regulate a diverse set of physiologic functions. The type A1 receptors inhibit adenylyl cyclase, and play a role in the fertilization process. Animal studies also suggest a role for A1 receptors in kidney function and ethanol intoxication. Transcript variants with alternative splicing in the 5' UTR have been found for this gene. [provided by RefSeq, Jul 2008]
