Principle of the Assay: C6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-C6 antibody and an C6-HRP conjugate. The assay sample and buffer are incubated together with C6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C6 concentration since C6 from samples and C6-HRP conjugate compete for the anti-C6 antibody binding site. Since the number of sites is limited, as more sites are occupied by C6 from the sample, fewer sites are left to bind C6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C6 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a component of the complement cascade. The encoded protein is part of the membrane attack complex that can be incorporated into the cell membrane and cause cell lysis. Mutations in this gene are associated with complement component-6 deficiency. Transcript variants encoding the same protein have been described.[provided by RefSeq, Nov 2012]
Uniprot Description
C6: Constituent of the membrane attack complex (MAC) that plays a key role in the innate and adaptive immune response by forming pores in the plasma membrane of target cells. Defects in C6 are the cause of complement component 6 deficiency (C6D). A rare defect of the complement classical pathway associated with susceptibility to severe recurrent infections, predominantly by Neisseria gonorrhoeae or Neisseria meningitidis. Belongs to the complement C6/C7/C8/C9 family.
Protein type: Secreted; Secreted, signal peptide
Chromosomal Location of Human Ortholog: 5p13
Cellular Component: membrane attack complex; extracellular region

Product Notes
The Rabbit C6 c6 (Catalog #AAA7239738) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7239738 ELISA Kit recognizes Rabbit C6. It is sometimes possible for the material contained within the vial of "Complement component C6 (C6), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
