Principle of the Assay: The LumiAb Human PAI-1 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human PAI-1 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human PAI-1 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human PAI-1 or Plasminogen Activator Inhibitor 1, also known as Serpin Peptidase Inhibitor Clade E, is a 402 amino acid cytokine protein that acts as "bait" for tissue plasminogen activator, urokinase and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. PAI-1 is expressed from the SERPINE1 gene located at locus 7q21.3-7q22 on chromosome 7. After initial synthesis, the 23 residue signal sequence is removed, allowing for the proper folding and maturation of the PAI-1 protein. Defects in SERPINE1 are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1D), a hematologic disorder characterized by increased bleeding after trauma, injury, or surgery; affected females have menorrhagia. The bleeding defect is due to increased fibrinolysis of fibrin blood clots due to deficiency of plasminogen activator inhibitor-1, which inhibits tissue and urinary activators of plasminogen. Moreover, high concentrations of PAI-1 seem to contribute to the development of venous but not arterial occlusions. PAI-1 is typically found in plasma and platelets as well as in endothelial, hepatoma and fibrosarcoma cells. A usual post-translational modification that occurs is the inactivation of PAI-1 by proteolytic attack of the urokinase-type (u-PA) and the tissue-type (TPA), cleaving Arg369-Met370 peptide bond.

NCBI Description

This gene encodes a member of the pentraxin protein family. The expression of this protein is induced by inflammatory cytokines in response to inflammatory stimuli in several mesenchymal and epithelial cell types, particularly endothelial cells and mononuclear phagocytes. The protein promotes fibrocyte differentiation and is involved in regulating inflammation and complement activation. It also plays a role in angiogenesis and tissue remodeling.
