Related Product Information for anti-WNT3A antibody
The Wnt family of consists of ligands involved in cell development, fate determination, organogenesis and oncogenesis. Information on Wnt signaling has been inferred from genetic analysis of wingless (wg) signaling in Drosophila. The Wnt signaling cascade initiates at the cell membrane when Wnt interacts with its receptor, a member of the Frizzled (Fz) transmembrane receptor family and lipoprotein receptor-related proteins 5 and 6 (LRP-5/6). This interaction relays the signal into the interior of the cell where disheveled (dsh) is brought to the membrane and gets phosphorylated/activated. Dsh promotes the inactivation of GSK3 kinase activity, thus preventing the phosphorylation of beta-Catenin and allowing it to accumulate as it avoids ubiquination and subsequent proteosome degradation. Stable beta-Catenin accumulates in the cytoplasm where it interacts with lymphoid enhancer factor/T cell factor (LEF/TCF) and translocates into the nucleus. This results in activation of TCF-responsive genes which are thought to play key roles in development and cancer. Negative regulators of the pathway may include axin and the tumor suppressor APC.

NCBI Description

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 96% amino acid identity to mouse Wnt3A protein, and 84% to human WNT3 protein, another WNT gene product. This gene is clustered with WNT14 gene, another family member, in chromosome 1q42 region. [provided by RefSeq]

Uniprot Description

WNT3A: Ligand for members of the frizzled family of seven transmembrane receptors. Wnt-3 and Wnt-3a play distinct roles in cell-cell signaling during morphogenesis of the developing neural tube. Interacts with PORCN. Interacts with APCDD1 and WLS. Component of the Wnt-Fzd-LRP5-LRP6 signaling complex that contains a WNT protein, a FZD protein and LRP5 or LRP6. Interacts directly in the complex with LRP6. Moderately expressed in placenta and at low levels in adult lung, spleen, and prostate. Belongs to the Wnt family.

Product Notes

The WNT3A wnt3a (Catalog #AAA622480) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Wnt3a (Wingless-type MMTV (Mouse Mammary Tumor Virus) Integration Site Family Member) reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Wnt3a can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in Immunohistochemistry and Western Blot. Dilution: Immunohistochemistry (Paraffin): 1:200 stains whole-mount transverse section of E12.5 mouse embryos. Incubated with diluted W3011-02B for 1 hour at RT. Tissue was pretreated with no heat induced epitope retrieval, citrate pH 6. Western Blot: 1:1000. Rat whole brain lysate was resolved by electrophoresis and transferred to PVDF. Proteins were visualized using an IgG (HRP) Dk xRb and a chemiluminescence detection system. Researchers should empirically determine the suitability of the WNT3A wnt3a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Wnt3a, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
