Specificity
This item specifically recognises VPS34 (vacuolar protein sorting 34), otherwise known as PI3-kinase type 3 (PIK3C3), a ubiquitously expressed class III PI3K (phosphoinositide 3-kinase), and member of the PI3/PI4-kinase family, which specifically utilizes phosphatidylinositol (PtdIn) as a substrate, and functions as the catalytic subunit of the PI3K complex.
VPS34 plays a role in the transport of lysosomal enzyme precursors to lysosomes, and in endocytosis and vesicular trafficking, and has been strongly implicated as a regulator of autophagy, forming a multi-protein complex with the proautophagic tumor suppressors Beclin1/Atg6, Bif-1, and UVRAG, in the initiation of autophagosome formation. Distinct complexes of VPS34 have been shown to regulate endocytic processes vital for late-stage autophagosome-lysosome fusion.
This item has been tested in immunofluorescence using HUVEC cells, in conjunction with Hoescht 33342 (blue) nuclear staining.

Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Involved in the transport of lysosomal enzyme precursors to lysosomes. Required for the abcission step in cytokinesis. Required for transport from early to late endosomes. Heterodimer. This subunit, part of a complex composed of regulatory and catalytic subunits, associates with regulatory subunit PIK3R4. Forms a complex with BECN1, PIK3R4 and either UVRAG and KIAA0226/Rubicon, or with ATG14. In this complex, presence of UVRAG and ATG14 are mutually exclusive. Part of a complex composed of PIK3R4 and PIK3CB. Interacts with RAB7A in the presence of PIK3R4. Ubiquitously expressed, with a highest expression in skeletal muscle. Belongs to the PI3/PI4-kinase family.
