Preparation and Storage
This antibody can be stored at 2-8 degree C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 degree C to -80 degree C. Avoid repeated freeze-thaw cycles.

Immunofluorescence (IF)

(Immunofluorescence staining of SUPT16H in U2OS cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human SUPT16H polyclonal antibody (dilution ratio 1:100) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus.)

The histone chaperone FACT (facilitates chromatin transcription) is a large heterodimeric complex with roles in transcription, replication, and repair. FACT promotes and subsequently restricts access to DNA as a result of dynamic nucleosome reorganization. The human FACT complex is composed of two polypeptides identified as Spt16 (Suppressor of ty 16) and SSRP1 (structure-specific recognition protein 1) that are both essential for nucleosome reorganization. The Spt16 protein is divided into three distinct structural domains and a highly acidic intrinsically disordered C-terminal domain required for alleviation of nucleosome blockage during polymerase II elongation. The SSRP1 component of FACT is divided into five well-defined domains, which includes a C-terminal HMG-1 DNA binding domain.
