Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Serpin A9 (Serine proteinase inhibitor-clade A #9; also GCET1, Centerin and Serpin A11) is a 46 kD, secreted member of the a1-antitrypsin (clade A)-subfamily, serpin superfamily of protease inhibitors. It is only produced by normal and lymphoma germinal center B cells. Its function is unknown, but it presumably acts as an inhibitor of a trypsin-like protease. Mature human Serpin A9 is presumably 394aa in length. It contains one Serpin motif (aa405-415). There are multiple splice variants associated with Serpin A9. There are alternate start sites at Met19, Met99 and Met150. A fourth isoform shows a two aa substitution for (aa369-435), while a fifth isoform shares this same substitution coupled with a 39aa substitution for (aa1-75). A sixth variant shows a deletion of (aa91-190). Mature human Serpin A9 shares 67% aa identity with mouse Serpin A9.

NCBI Description

The superfamily of high molecular weight serine proteinase inhibitors (serpins) regulate a diverse set of intracellular and extracellular processes such as complement activation, fibrinolysis, coagulation, cellular differentiation, tumor suppression, apoptosis, and cell migration. Serpins are characterized by well-conserved a tertiary structure that consists of 3 beta sheets and 8 or 9 alpha helices (Huber and Carrell, 1989 [PubMed 2690952]). A critical portion of the molecule, the reactive center loop connects beta sheets A and C. Protease inhibitor-8 (PI8; SERPINB8) is a member of the ov-serpin subfamily, which, relative to the archetypal serpin PI1 (MIM 107400), is characterized by a high degree of homology to chicken ovalbumin, lack of N- and C-terminal extensions, absence of a signal peptide, and a serine rather than an asparagine residue at the penultimate position (summary by Bartuski et al., 1997 [PubMed 9268635]).[supplied by OMIM, Jan 2010]
