Function
Functions as a calcium- and diacylglycerol(DAG)- regulated nucleotide exchange factorspecifically activating Ras through theexchange of bound GDP for GTP (PubMed: 15899849,PubMed: 23908768). Activatesthe Erk/MAP kinase cascade (PubMed: 15899849). RegulatesT-cell/B-cell development, homeostasis anddifferentiation by coupling T-lymphocyte/B-lymphocyte antigen receptors to Ras(PubMed: 10807788, PubMed: 12839994).Regulates NK cell cytotoxicity and ITAM-dependent cytokine production by activation of Ras-mediated ERK and JNK pathways (PubMed: 19933860). Functions in mast cell degranulation and cytokinesecretion, regulating FcERI-evoked allergic responses (By similarity). May also function in differentiation of other cell types (PubMed: 12845332).
Cellular Location
Cytoplasm, cytosol. Cell membrane; Peripheral membrane protein. Golgiapparatus membrane; Peripheral membraneprotein. Endoplasmic reticulum membrane;Peripheral membrane protein. Note=Foundboth in the cytosol and associated with membranes. Relocalization to the cellmembrane upon activation is F-actin-dependent. Translocates to the Golgiin response to phorbol ester or nerve growth factor. Localizes to somata and dendrites but not to axons of hippocampal pyramidal cells (By similarity).
Tissue Location
Expressed in brain with higher expression incerebellum, cerebral cortex and amygdala. Expressed in the hematopoietic system. Expressed in T-cells (at protein level)Expressed in NK cells (at protein level)(PubMed:19933860)
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 2 weeks. For long term storage store at -20 degree C in small aliquots to prevent freeze-thaw cycles.

NCBI Description

This gene is a member of a family of genes characterized by the presence of a Ras superfamily guanine nucleotide exchange factor (GEF) domain. It functions as a diacylglycerol (DAG)-regulated nucleotide exchange factor specifically activating Ras through the exchange of bound GDP for GTP. It activates the Erk/MAP kinase cascade and regulates T-cells and B-cells development, homeostasis and differentiation. Alternatively spliced transcript variants encoding different isoforms have been identified. Altered expression of the different isoforms of this protein may be a cause of susceptibility to systemic lupus erythematosus (SLE). [provided by RefSeq, Jul 2008]

Uniprot Description

RASGRP1: Functions as a diacylglycerol (DAG)-regulated nucleotide exchange factor specifically activating Ras through the exchange of bound GDP for GTP. Activates the Erk/MAP kinase cascade. Couples T-lymphocytes and B-lymphocytes antigen receptors to the activation of Ras. Hence, regulates T-cells and B-cells development, homeostasis and differentiation. Functions also in FcERI-evoked degranulation and cytokine secretion by mast cells, regulating allergic responses. May also function in other cell types differentiation. Defects in RASGRP1 may contribute to susceptibility to systemic lupus erythematosus (SLE). SLE is a chronic, inflammatory and often febrile multisystemic disorder of connective tissue. It affects principally the skin, joints, kidneys and serosal membranes. SLE is thought to represent a failure of the regulatory mechanisms of the autoimmune system. Aberrantly spliced isoforms and/or diminished levels of RASGRP1 are found in a cohort of SLE patients raising the possibility that dysregulation of this signaling protein contributes to the development of autoimmunity in a subset of SLE patients. Belongs to the RASGRP family. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The RASGRP1 rasgrp1 (Catalog #AAA9206589) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 495-521. The RASGRP1 Antibody (Center) reacts with Human (Predicted Reactivity: Mouse, Rat) and may cross-react with other species as described in the data sheet. AAA Biotech's RASGRP1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunohistochemistry (IHC), Flow Cytometry (FC/FACS). WB~~1:1000 IHC-P~~1:10~50 FC~~1:10~50. Researchers should empirically determine the suitability of the RASGRP1 rasgrp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RASGRP1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
