Host
Rabbit
Reactivity
Mouse, rat
Clonality
Polyclonal
Specificity
Specific for the ~140k NR2C subunit of the NMDA receptor phosphorylated at Ser1244. Immunolabeling is blocked by preadsorption of antibody with the phospho-peptide that was used to generate the antibody but not by the corresponding dephospho-peptide.
Purity/Purification
Affinity Purified (Prepared from rabbit serum by affinity purification via sequential chromatography on phospho- and dephosphopeptide affinity columns.)

Sequence Length
1250
Applicable Applications for anti-GRIN2C antibody
Western Blot (WB)
Application Notes
Quality Control: Western blots performed on each lot.
WB: 1:1000
Antigen
Synthetic phospho-peptide surrounding the phospho Ser1244 of the NR2C-subunit of rat NMDA receptor.
Immunogen Information
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser1244 conjugated to KLH
Immunogen Species
Rat
Species Reactivity Note
The antibody has been directly tested for reactivity in Western blots with mouse and rat tissue. It is also expected that the antibody will react with bovine, canine, human and non-human primates as these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance
The ion channels activated by glutamate that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR). The NMDAR plays an essential role in memory, neuronal development and it has also been implicated in several disorders of the central nervous system including Alzheimer's, epilepsy and ischemic neuronal cell death (Grosshans et al., 2002; Wenthold et al., 2003; Carroll and Zukin, 2002). The NMDA receptor is also one of the principal molecular targets for alcohol in the CNS (Lovinger et al., 1989; Alvestad et al., 2003; Snell et al., 1996). The NMDAR is also potentiated by protein phosphorylation (Lu et al., 1999). The NR2C subunit of the receptor is thought to influence the NMDAR conductance level (Ebralidze et al., 1996). Phosphorylation of Ser1244 has been shown to regulate NMDA receptor channel function (Chen et al., 2006).
Preparation and Storage
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.

NCBI Description

subunit that in combination with NMDAR1 displays NMDA receptor activity; may contribute to NMDA receptor functional diversity [RGD, Feb 2006]

Uniprot Description

Function: NMDA receptor subtype of glutamate-gated ion channels possesses high calcium permeability and voltage-dependent sensitivity to magnesium. Mediated by glycine.

Subunit structure: Forms heteromeric channel of a zeta subunit (GRIN1), a epsilon subunit (GRIN2A, GRIN2B, GRIN2C or GRIN2D) and a third subunit (GRIN3A or GRIN3B). Interacts with PDZ domains of INADL and DLG4. Interacts (via PDZ-binding motif) with SNX27 (via PDZ domain); the interaction is required for recycling to the plasma membrane when endocytosed and prevent degradation in lysosomes. Ref.3 Ref.4

Subcellular location: Cell membrane; Multi-pass membrane protein. Cell junction › synapse › postsynaptic cell membrane; Multi-pass membrane protein.

Sequence similarities: Belongs to the glutamate-gated ion channel (TC 1.A.10.1) family. NR2C/GRIN2C subfamily. [View classification]

Sequence caution: The sequence BAA02499.1 differs from that shown. Reason: Erroneous initiation.

Product Notes

The GRIN2C grin2c (Catalog #AAA502150) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Phospho Ser1244 NMDA Receptor, NR2C Subunit reacts with Mouse, rat and may cross-react with other species as described in the data sheet. AAA Biotech's NMDA NR2C Subunit (Ser1244) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Quality Control: Western blots performed on each lot. WB: 1:1000. Researchers should empirically determine the suitability of the GRIN2C grin2c for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NMDA NR2C Subunit (Ser1244), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
