Western Blot (WB)
(Western blot analysis of extracts from 3T3, using NLRC3 Antibody. The lane on the left was treated with blocking peptide.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612170 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612170 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Subcellular Location: Cytoplasm.
Subunit Structure: Directly interacts (via CARD) with TMEM173/STING; this interaction reduces TMEM173 trafficking to the perinuclear region in response to interferon stimulatory DNA. Also interacts, but to a lesser extent, with TBK1 (PubMed:24560620). Interacts with TRAF6; this interaction results in decreased TRAF6 'Lys-63'-linked polyubiquitination, but leaves 'Lys-48'-linked chains unchanged, promoting TRAF6 protein degradation (PubMed:22863753). Interacts with PIK3R1/PIK3R2; this interaction disrupts the association between PIK3R1/PIK3R2 and the p110 catalytic subunit PIK3CA/PIK3CB/PIK3CD and reduces PIK3R1/PIK3R2 activation (By similarity). Weakly interacts with PYCARD/ASC. Interacts with CASP1 and CASP5 (PubMed:25277106).
Similarity: The leucine-rich repeat domain may reduce the interaction with TMEM173/STING.Belongs to the NLRP family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)115kDa.
NCBI Description
This gene encodes a NOD-like receptor family member. The encoded protein is a cytosolic regulator of innate immunity. This protein directly interacts with stimulator of interferon genes (STING), to prevent its proper trafficking, resulting in disruption of STING-dependent activation of the innate immune response. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2014]
Uniprot Description
NLRC3: May modulate T-cell activation. Decreases the transcription of genes that are normally up-regulated after T-cell stimulation. Delays degradation of NFKBIA/IKBA. Belongs to the NLRP family. 4 isoforms of the human protein are produced by alternative splicing.
Chromosomal Location of Human Ortholog: 16p13.3
Cellular Component: cytoplasm
Molecular Function: ATP binding
Biological Process: I-kappaB kinase/NF-kappaB cascade; T cell activation; regulation of protein ubiquitination; inhibition of NF-kappaB transcription factor; response to lipopolysaccharide; negative regulation of I-kappaB kinase/NF-kappaB cascade

Product Notes
The NLRC3 nlrc3 (Catalog #AAA9612170) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The NLRC3 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's NLRC3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the NLRC3 nlrc3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NLRC3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
