This antibody is intended for use in Immunohistochemical applications on formalinfixed paraffin-embedded tissues (FFPE), frozen tissue sections and cell preparations. Interpretation of results should be performed by a qualified medical professional.

Precautions
1. For profesional users only. Ensure results are interpreted by a medical professional.
2. This product contains sodium azide (NaN3), a toxic chemical which may react with plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent sodium azide build-up.
3. Ensure proper handling procedures are used with reagent. Always wear proper laboratory equipment such as laboratory coat and gloves when handling reagents.
4. Unused solution should be disposed of according to local and federal regulations.
5. Do not ingest reagent. If reagent ingested, contact a poison control center immediately.
6. For complete recommendations for handling biological specimens please refer to the CDC document, “Guidelines for Safe Work Practices in Human and Animal Medical Laboratories” (3).
Specimen Preparation
Paraffin sections: The antibody can be used on formalin-fixed paraffin-embedded (FFPE) tissue sections. Ensure tissue undergoes appropriate fixation to ensure best results.

Frozen sections and cell preparations: The antibody can be used for labeling acetone-fixed frozen sections and acetone-fixed cell preparations.

Preparation and Storage

Myeloperoxidase (MPO) is a peroxidase enzyme most abundantly present in neutrophil granulocytes. It is a lysosomal protein stored in azurophilic granules of the neutrophil. MPO has a heme pigment, which causes its green color in secretions rich in neutrophils, such as pus and some forms of mucus. Historically, immunohistochemical staining for myeloperoxidase was used in the identification of Acute Myeloid Leukemia to demonstrate that the leukemic cells were derived from the myeloid lineage. Myeloperoxidase staining is still important in the identification of Extramedullary Leukemia or Chloroma. Myeloperoxidase detects granulocytes and monocytes in blood and precursors of granulocytes in the bone marrow. This antibody can detect myeloid cell populations of the bone marrow as well as in other sites.

NCBI Description

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils. [provided by RefSeq]

Function: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.

Catalytic activity: Cl- + H2O2 + H+ = HClO + H2O.Cl- + H2O2 = HOCl + 2 H2O.

Cofactor: Binds 1 calcium ion per monomer.Binds 1 heme B (iron-protoporphyrin IX) group covalently per monomer.

Subunit structure: Homodimer; disulfide-linked. Each monomer consists of a light and a heavy chain. Ref.13 Ref.20

Subcellular location: Lysosome.

Involvement in disease: Defects in MPO are the cause of myeloperoxidase deficiency (MPD) [

MIM:254600]. MPD is an autosomal recessive defect that results in disseminated candidiasis. Ref.23 Ref.24 Ref.25 Ref.26 Ref.27

Sequence similarities: Belongs to the peroxidase family. XPO subfamily.
