Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The mammalian target of Rapamycin, mTOR, also know as FRAP or RAFT (1-3), is a serine/ threonine protein kinase. mTOR acts as a sensor for ATP, amino acids, balancing the availability of nutrients and cell growth (4,5). When sufficient nutrients are available, mediated by phosphatidic acid (6), mTOR transmits a positive signal to p70 S6 kinase and participates in the inactivation of the eIF4E inhibitor 4E-BP1. These events result in activation of the translation of specific mRNA subpopulations. mTOR is phosphorylated at serine 2448 via the PI3 kinase/Akt signaling pathway and autophosphorylated at serine 2481 (6,7).
