Relevance: The mammalian matrix metalloproteinases (MMPs) degrade extracellular matrix in physiological and pathological processes. Of the sixteen members of matrix metalloproteinase family ten exist in soluble form whereas MT MMPs exist as integral membrane proteins. MMP15, first described as MTMMP2, is a metalloproteinase with a type I transmembrane domain. Initially reported in lung tissue, MMP15 message was also seen in testis, intestines, colon, heart, liver, kidney, pancreas and muscle. Western blots showed bands at 72 kD, 48 kD and 44 kD. Subsequent publications have focused on message, and found MMP15 in a wide range of tumor cell lines, as well as the synovial membrane from patients with rheumatoid arthritis. MMP15 is usually found in much lower levels than the more ubiquitous MMP14 (MT1MMP). MMP15 may be shed into the culture media when activated, but it is unclear if the shed form is proteolytically active. Cell lysates are the best source for Western blotting of MMP15.

RP1MMP15 works well for Western Blotting applications, and when used against the reduced protein identifies bands at 72 Kd and 55 Kd as well as activation/breakdown product at 48 kD. Often the MMP-15 is fully activated, and the 48 kD band is the dominant band seen. MMP-15 may be shed into the culture media when activated, but it is unclear if the shed form is proteolytically active. Cell lysates are the best source for Western blotting of MMP-15. The antibody can recognize native or reduced MT2MMP, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples. Researchers should empirically determine the suitability of the Membrane-Type Matrix Metalloproteinase-2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
