This antibody binds to the propeptide domain of MMP24. It recognizes only the zymogen of MMP24, and not the activated forms. It can recognize native or reduced forms. The antibody does not cross react with the other MMP family members (MMP1, MMP2, MMP3, MMP9).

Application Notes
The antibody works well for Western Blotting applications, and when used against the reduced protein identifies bands at 65 Kd. The antibody recognizes only the zymogen of MMP-24, and not the activated forms. The antibody recognizes human and murine MMP-24. The antibody can recognize native or reduced MT5MMP, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. MT5MMP has been reported to be elevated in several tumor cell lines, and is constituitively produced by some normal cell lines. MMP-24 is also found in brain tissue, along with other MT-MMP enzymes. Treatment of cells with Concanavolin-A or the phorbol ester TPA stimulates production of MT5MMP in some cell types, and the enzyme can be recovered in cell lysates. Shed forms of MT5MMP have also been reported. A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples.

Preparation and Storage
This product is stable for several weeks at 4 degree C as an undiluted liquid. Dilute only prior to immediate use.
For extended storage, aliquot contents and freeze at-20 degree C or below. Avoid cycles of freezing and thawing. Expiration date is one (1) year from date of receipt.

Description: The antibody is a Rabbit antibody to the membrane-bound Matrix Metalloproteinase-5 (MMP-24), made with a synthetic peptide based on the mouse sequence. The antibody binds to the Propeptide domain of MT5MMP. The antibody binds to MT5MMP, but does not cross react with the other MMP family members (MMP1, MMP2, MMP3, MMP-9). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative, and 50% glycerol as cryoprotectant.

Relevance: Matrix Metalloproteinase 24 (MMP24, MT5MMP) has been described as a proteoglycanase, which is critical in the turnover of ECM (extracellular matrix) components in the brain. MMP24 contains a candidate signal sequence, a propeptide region, a potential furin recognition motif, a zinc-binding catalytic domain, a hemopexin-like domain, a 24-residue hydrophobic region, which acts as a potential transmembrane domain, and a short cytosolic domain. Unlike other MTMMPs, MMP24 (MT5MMP) sheds from the cell surface as soluble proteinases, allowing its versatility as both a cell bound and soluble proteinase in ECM remodeling processes. MMP24 is elevated in several tumor cell lines, and is also produced by some normal cell lines. It is predominantly expressed in the brain, indicating a role in nervous system development. MMP24 is also highly expressed during embryonic development. The mouse MMP24 gene maps to chromosome 2.
